I'd recommend doing the 2-ply separately. Problem is how to apply the glue evenly to just the veneer if you're trying to do it all at once, as it does start to curl as soon as glue is applied. Plus I'd think that much more difficult to keep things lined up with an extra layer with wet glue slipping around. Darryl's trick that I now use is to make the underlayer of the 2-ply slightly larger than the show veneer, so that it's edges can be taped down to the bottom caul (I use packing tape). This greatly aids in keeping it down flat while rolling the glue onto it.
As far as leaving it overnight before pressing, that shouldn't be a problem. Just don't leave the 2-ply "out" to cure, must leave it under a caul. Otherwise will curl up as the glue dries, and I find is extremely vulnerable to cracking when then trying to unroll it. But if left under a caul, it is still flexible enough to apply to curved forms.
Personally, I don't like leaving veneer tape between the 2 layers either. Definitely weaker than just wood on wood, and I don't like to take any chances on the show veneer. So I use 2-hole tape on the underside of the backing veneer, and then remove it before gluing the completed 2-ply to the substrate.
Actually I just don't much like 3 hole tape, as I find it disintegrates so easily just trying to get it off the veneer tape dispenser once wet. It just doesn't seem to hold a seam that well either, especially if the veneer is anything but dead flat.
